SFONDO
Il gene 1 indotto da tazarotene (TIG1) è un gene soppressore tumorale di tipo II inducibile dai retinoidi. L’isoforma B di TIG1 (TIG1B) inibisce la crescita e l’invasione delle cellule tumorali. L’espressione di TIG1B è spesso sottoregolata in vari tessuti tumorali; tuttavia, l’espressione e le attività dell’isoforma TIG1A devono ancora essere riportate. Pertanto, questo studio ha esaminato gli effetti delle isoforme TIG1A e TIG1B sulla crescita cellulare e sui profili di espressione genica utilizzando cellule di cancro del colon.
METODI
I cloni stabili TIG1A e TIG1B derivati da cellule di cancro al colon HCT116 e SW620 sono stati stabiliti utilizzando il sistema GeneSwitch; L’espressione dell’isoforma TIG1 è stata indotta dal trattamento con mifepristone. La crescita cellulare è stata valutata utilizzando i test di proliferazione cellulare e formazione di colonie WST-1. L’interferenza dell’RNA è stata utilizzata per esaminare i cambiamenti di mediazione di TIG1 nella crescita cellulare. I profili di espressione genica sono stati determinati utilizzando microarray e convalidati utilizzando la reazione a catena della polimerasi in tempo reale e analisi Western blot.
RISULTATI
Entrambe le isoforme di TIG1 sono state espresse a livelli elevati nei tessuti normali della prostata e del colon e sono state sottoregolate nelle linee cellulari di cancro del colon. Entrambe le isoforme TIG1 hanno inibito in modo significativo la crescita di cellule HCT116 transitoriamente trasfettate ed esprimendo stabilmente cellule TIG1A e TIG1B HCT116 e SW620. L’espressione dei geni 129 e 55 è stata alterata all’induzione dell’espressione di TIG1A e TIG1B, rispettivamente, nelle cellule HCT116 che esprimono stabilmente. Dei geni analizzati, 23 e 6 geni erano rispettivamente sovraregolati e sottoregolati nelle cellule che esprimono TIG1A e TIG1B. La sovraregolazione della chinasi 5 del recettore accoppiato alla proteina G (GRK5) è stata confermata utilizzando la reazione a catena della polimerasi in tempo reale e analisi Western blot in entrambe le linee cellulari stabili TIG1. Il silenziamento dell’espressione di TIG1A o GRK5 ha ridotto significativamente la soppressione della crescita cellulare mediata da TIG1A.
CONCLUSIONI
L’espressione di entrambe le isoforme di TIG1 è stata osservata nei tessuti normali della prostata e del colon ed è stata sottoregolata nelle linee cellulari di cancro del colon. Entrambe le isoforme TIG1 hanno soppresso la crescita cellulare e stimolato l’espressione di GRK5 nelle cellule HCT116 e SW620. Il knockdown dell’espressione di GRK5 ha alleviato la soppressione della crescita indotta da TIG1A delle cellule HCT116, suggerendo che GRK5 media la soppressione della crescita cellulare da parte di TIG1A. Pertanto, TIG1 può partecipare alla sottoregolazione della segnalazione accoppiata alla proteina G sovraregolando l’espressione di GRK5.
Metodi spettrofotometrici che indicano la stabilità per la determinazione del tazarotene in presenza del suo prodotto di degradazione alcalino mediante tecniche spettrofotometriche derivate.
È stata studiata la stabilità del tazarotene (TZ) e sono stati utilizzati due metodi di indicazione della stabilità, vale a dire, il derivato primo e un metodo spettrofotometrico del rapporto dei derivati, per determinare il tazarotene in presenza del suo prodotto di degradazione alcalina (HD) utilizzando metanolo come solvente. È stata ottenuta una relazione lineare nell’intervallo 1-10 µg ml ¹ per entrambi i metodi.
Applicando i metodi proposti, è stato possibile determinare il tazarotene nella sua polvere pura da con precisione 99,35 ± 1,410 (n = 10) per il metodo della derivata prima e 99,45 ± 1,053 (n = 10) per il metodo del rapporto dei derivati. I metodi del derivato primo e del rapporto dei derivati sono stati utilizzati per l’analisi di miscele preparate in laboratorio contenenti diversi rapporti di tazarotene e del suo prodotto di degradazione ed erano validi in presenza rispettivamente fino al 70% e all’80% di prodotto di degradazione. I metodi proposti sono stati convalidati e trovati adatti come metodi di analisi indicanti la stabilità per il tazarotene in formulazioni farmaceutiche.
[Retinoidi: meccanismi d’azione].
I retinoidi, derivati della vitamina A, sono molecole naturali o sintetiche con effetti pleiotropici, che regolano il differenziamento, la proliferazione e l’apoptosi cellulare. Nella cellula bersaglio, i metaboliti naturali attivi acido retinoico (RA) e acido 9-cis-retinoico sono sintetizzati dal retinolo mediante un processo in due fasi con il metabolita intermedio retinaldeide. Nel 1987, l’identificazione dei recettori nucleari dell’acido retinoico che appartengono alla superfamiglia dei recettori nucleari ha portato a un progresso significativo nella comprensione del meccanismo d’azione dei retinoidi. Esistono due famiglie di recettori nucleari retinoidi (RNR), i recettori RA (RAR), il cui ligando naturale è RA, e i recettori X retinoidi (RXR), il cui ligando naturale è acido 9-cis-retinoico. Tra i retinoidi sintetici, isotretinoina, acitretina, tazarotene e adapalene sono ligandi del RAR, il bexarotene è il primo rexinoide (ligando dell’RXR), l’alitretinoina il primo panagonista (RAR + RXR).

Description: Tissue factor is a transmembrane glycoprotein of the cytokine receptor superfamily that acts as a receptor for coagulation factor VII (fVII) to trigger initiation of the coagulation cascade in response to vascular injury. Expression of tissue factor occurs constitutively within most extravascular and perivascular cells and at high levels within critical organs and tissue. Tissue factor is not normally expressed freely on the surface of circulating blood cells due to its pro-coagulant effect, but is instead stored on the surface of mononuclear and endothelial cells in microparticles that can shed into circulation in response to vascular injury, pro-inflammatory cytokines, or microbial ligands. Tissue factor can also be shed into circulation by cancer cells where its expression in a number of cancer types has been linked to tumor progression, metastatic potential, thrombosis, and angiogenesis. Expression of tissue factor has been shown to be inducible by select cytokines in a number of cell types, including IL-1β and TNF-α in vascular endothelial cells and macrophages, and TNF-α, IL-6, and FGF-Basic in monocytes, among others. CHO cell-derived Recombinant Human Tissue Factor is a glycoprotein homodimer comprised of the 214-amino-acid length monomer containing a C-terminal His-Tag. It has a calculated molecular weight of approximately 24.4 kDa, and as a result of glycosylation, Recombinant Human Tissue Factor migrates with an apparent molecular mass of approximately 45-55 kDa by SDS-PAGE gel, under reducing conditions. |

Per regolare l’espressione dei geni bersaglio, NRR deve dimerizzare. RXR sono obbligatori nei dimeri (eterodimeri RAR-RXR, omodimeri RXR-RXR). I dimeri si legano a specifiche sequenze di DNA, presenti nei promotori dei geni bersaglio. Quando il ligando, naturale o sintetico, si lega all’RNR, vengono reclutati i coattivatori e vengono attivati i fattori di trascrizione. Nella cellula bersaglio, i retinoidi non utilizzati vengono degradati nei metaboliti polari dagli enzimi del citocromo P450.
